Q4: In the COA of your company's aluminum hydroxide adjuvant, there is an indicator that Al2O3 is about 2%. Is Al2O3 the main component in this product?
The specific components in this product are not convenient to disclose. The Al2O3 index in the COA is also a converted index based on the relevant components of the specific product. There is also an Al3+ conversion index in the COA, which is approximately 10mg/ml. This Al3+ conversion index is the main reference index for customers when using it
Q5: Does the aluminum adjuvant need emulsification?
Aluminum salt adjuvants are not oily adjuvants and belong to inorganic materials, so there is no concept of emulsification.
Q6: My aluminum adjuvant has precipitated and cannot be mixed evenly. What is the reason?
Under normal circumstances, shaking it up and down can mix it well. Our adjuvant has also been optimized and adjusted in terms of suspension stability. After mixing, it can generally remain in a homogeneous state for more than several hours. However, please note that after preparing the vaccine, it should not be left for a long time. Vaccines that have been left for a long time need to be shaken again to make them uniform before use. In addition, please note that our aluminum salt adjuvant must not be frozen. Freezing will affect the structure of the aluminum salt adjuvant, leading to irreversible aluminum aggregation and even serious caking problems. Uneven temperatures in some refrigerators can also cause local freezing of adjuvants, thereby affecting product quality. It is recommended to place it near the door of the refrigerator to maintain a low temperature.
Q7: I left my adjuvant at room temperature. Does it have any effect?
There is no effect. The adjuvant can be stored at room temperature (25℃)
Q8: How to mix the aluminum adjuvant and antigen? Do I need to prepare at a 1:1 volume ratio like other adjuvants?
The instruction manual provides reference dosage information. After mixing with the antigen, simply shake it evenly by physical methods such as manual shaking or pipetting with a pipette tip. Regarding the volume ratio of 1:1, if necessary, teachers can dilute it by themselves. For instance, normal saline is compatible. In vaccine development, researchers mainly focus on the content of aluminum adjuvants. Please adjust and optimize the content based on your own experimental conditions, literature, data, etc.
Q9: I calculated the dosage for mice to be very small. One vial of adjuvant could be used hundreds or thousands of times. Did I miscalculate?
Regarding the specifications and prices of aluminum adjuvants, we have provided preferential prices by referring to the market pricing. Due to significant differences in weight among different species, there will be considerable variations in the frequency of use.
Q10: Does this contain magnesium hydroxide?
Does not contain Mg2+
Q11: What is the particle size of the aluminum hydroxide adjuvant?
Micrometer level
Q12: Is it sterile or non-sterile?
Sterile aliquoted
Q13: For mouse modeling, we recommend a dose of 50~100ug (aluminum ion per time). Literature reports using 60ul of Thermo product 77161, which converts to an aluminum ion concentration of 0.8mg per time. There's a big difference between the two dosages. How to choose?
The dosage provided in the instruction manual is the reference dose required for antibody preparation, not the modeling dose. Please explore the appropriate dosage based on the literature. The processes and ratios of adjuvants from different brands vary, so the reference dosages provided by different brands also differ. The common doses in the literature can be used as a reference starting point. Please adjust according to the evaluation results and determine the most suitable mouse strain, sensitization interval, allergen type and dose, adjuvant type and dose, etc.
Q14: In literature, a common dose for Thermo's product is 2mg aluminum hydroxide in 50ul. Switching to Biodragon's, if I still need to inject 2mg aluminum hydroxide, how many ul are needed?
The following calculation methods can be referred to:
Thermo77161 is quantified at 40mg/ml of aluminium hydroxide (AI(OH) 3,78.004 g/mol), and after conversion, the concentration of aluminium ions (AI3*, 26.98g/mol) is 13.83mg/ml
② Aluminum ion content of Boaolong adjuvant :10mg/ml.
50 ulx13. 33 mg/mL; 10 mg/ml = 69.175 ul
Or, 2mgx26.98g/mol; 78.004 g/mol,; 10 mg/ml = 69.17 ul
That is, for 2mg of aluminium hydroxide, approximately 70 μ l of adjuvant is required
