Degradation of DNA(A), DNA+RNA(B)contaminants in cell paste (CP)and conditioned media(CM)generated from a bio-reactor. 2L of culture of CHO cell was harvested at day 10 and total cell number reached 2x109.Amount of PanNuclease were added: lane 1&4: 0U, lane 2 & 5: 1U, Lane 3&6: 0.1U to treat 2x107 cells(CP) in 50μl of the buffer(20mM Tris-HCl ,pH8.0, 1M Urea, 1% Triton X-100)or 500ml of conditioned media (CM)and incubated at room temperature for 25 min. DNA and RNA were extracted, then separated. |
